I ordered some tissue cultures and im new to them. When they arrive I plan to leave them in their little bag in my greenhouse cabinet for 2 days for them to get used to the temperature before then putting them in fluval stratum in 100% humidity. My question is if I plan to eventually put these guys in appropriate soil mixes, how can I transition them there from fluval stratum? Also, Ive seen people do 1:1 fluval stratum to perlite off the bat but Ive run out of perlite. Is JUST fluval statum ok? Do I need perlite at all whether be off the bat or to help with transitioning? I have pumice on hand btw, idk if that can substitute perlite if necessary.

My routine for TC is::: put them in the place I plan to acclimate them for 3-6 days. If they have few roots, you can go longer (if they hopefully still had IBA or 2-4-D present, etc.). After that, get a fungicide/moldicide solution ready. I use Growsafe diluted in Distilled water (DI). Then have more DI for rinsing. Spray the bags down with 70% alcohol and let them sit for a minute. Also spray down any tools, i use 1 or two sets of tweezers, to help remove any yellowing leaves, and push them into the media. Then cut a bag open. Rince the excess agar/gellen gum off (use gloves to avoid and hormones present). After removing it all under the tap, remove any bad leaves or roots. Then put the plantlets in the growsafe solution. For at least 5 mins. I usually occasionally stir them. Afterwards rinse them with DI. Then dip the roots in rooting powder (IBA dilution). And plant in moistened stratum (perlite or mix). Close up the mini greenhouse to keep 100% humidity. Leave it fir 2 weeks. Then you can crack the greenhouse slightly to allow acclimating. I check if more moisture is needed at this time. After 1-2more weeks, depending on how they are doing. Crack it more. And water with a heavily diluted fertilizer.

After a few more weeks you can remove the lids completely. It you see any issues of melting. You can use Wilt Stop to give them a way coating. To help with acclimating.

Pumice can sub in... but Most people mix stratum because it's expensive. You do not need to mix it at all.

Thank you SO much!!! I just wanna ask, is the rooting hormone very important? Should I hurry up and get it, or is it ok that I don’t have it? Also, I personally use a copper fungicide for my plants, but it’s very strong (i buy it pure) so I dilute it. As long as I dilute it enough, should it be ok for the tissue cultures? Or is copper in general harmful for such fragile little tissue cultures?

It depends where you get your TC, type of plant... basically what the roots will look like. A fair amount will do multiplication stage and then as long as the stems and leaf are to size will skimp on IBA. Oorrr the species of plant sucks at rooting. So you'll get some plantlets with zero roots. And especially with TC they need the boost. Most hardware store will have a rooting powder and most are IBA based. You can always compare the IBA % if there's options, the higher the better.

I would personally not use a copper on them. They have negative zero protective coating on. Worse case I'd use a diluted peroxide over the copper sulphite. I know other people use that, or other fungicides, but this one I've found to work 100% and be gentle. I think it runs like $24 for the small bottle. But you literally use a few drops per pint jar of distilled.

Both came from the same Thai seller. But philodendron are trash at rooting in TC. And it takes forever

Oh my, I got like 2 philodendrons TC that are part of my order😅, the rest are anthuriums. Most likely i’ll totally pick up some rooting powder now before they arrive. Is there such thing as too high of a percentage of IBA? Online I for example see one brand that has bottles with different percentages from 0.10% up to 1.60%. 1.60% seems too strong to me, is it? Is something more like 0.30% better? Oh and do I need to dilute it at all? If so, how (powder)? I apologize for so many questions! You’ve been such an amazing help!

Don't apologize, ask away. I've never had an issue with high IBA. Most over the counter, in the store are low %. I regularly use one that is 0.01% Prop Drops (liquid), Bonide Rooting powder 0.10% (powder, or DipnGrow 1.0% ( liquid) the dipngrow is the one I usually use on the TC. I also have straight IBA for TC culturing that gets diluted. I plan to just do my own dilution now, once these run out.

You won't need to dilute the over the counter. If it's the powder, just put some in a dish and dip the butter of the plant in and plant. If it's the liquid, put it in a small dish and dip, or put the plantlet over something and drip it onto the but of the plantlet, then plant

You were an AMAZING help!!! I think I’ve got enough understanding of what to do now! Thank you again, and have a wonderful, wonderful day!!!
